MSE seems a good replacement for anti-virus and other anti-malware scanners, including Windows Defender, and the reviews I have seen are very reasonable.
It is not a firewall, so you will still need one of those, either Windows own firewall enabled, or a 3rd party one.
Not all scanners find everything though, and in particular with spyware, you could use something like Malwarebytes' Anti-malware, or SUPERAntispyware, or both, to complement MSE. They are both free, but the free versions do not allow you to use the resident scanners, meaning they will not load in the background. But that is fine, they can be used as manual scanners, once a week or so.
MSE replaced Windows Defender, so that is why you don't need WD.
