My USB flash drive isn't working with my desktop....

Hi, I have my flash drive and it worked just fine with my regular use desktop, then unexpectedly it stopped working from one day to the next one.
I checked all the options on the OS and I don't seem to encounter an answer.
Also I want to make notice that I have formatted the drive in several different machines to try to see if that was the issue solver, but sadly no. It works with every other system, but not my PC, most likely just my OS, I have tried to use the same system while booting from an XP PRO X32 environment and it worked, but on my regular Vista Home Premium X64 does not at all, even my car stereo accepts the data transmissions!
It do make the sound when inserted proving it actually recognizes the USB Mass Storage device (I have uninstalled the device already on device manager and noticed which HUB worked), then discovered it seems to install the drivers and say that the drive it's ready for use, and then again nothing appears on My Computer.
Please any assistance that solves this could be awesome.
